I have an 12 x 6 x 0.5 plank of walnut wood that I’ve cut into 5 irregular pieces. I’m going to make a foamboard mold to hold the pieces, leaving gaps between the pieces that I will fill with resin. My question is, should I hot glue (or any other method) the wooden pieces down before pouring the resin into the gaps so they don’t float around? Will resin get under the wood if I hot glue it down? And is it easy enough to remove the piece afterwards if I do glue it down first?

No, don’t hot glue them down. Instead pour a shallow layer of resin first that will adhere to the wood, but not deep enough that they will float.
